摘要: Movement and distribution of nuclei in fungi have been shown to be dependent on cytoplasmic microtubules the microtubule-associated motor dynein. We isolated hundreds Neurospora crassa mutants, known as ropy, that are defective nuclear distribution. Three ro genes, ro-1, ro-3 ro-4, encode subunits either dynein or activator complex, dynactin. In this report, we describe isolation initial characterization two additional ro-10 ro-11. ro-11 non-essential genes novel 24 kDa 75 kDa proteins respectively. Both mutants retain ability generate long microtubule tracks, suggesting defect is not caused by a gross cytoskeleton. RO10, well RO4 (actin-related protein ARP1, most abundant subunit dynactin), appears required for stability RO3 (p150Glued), largest propose lack proper distribution, because RO10 dynactin activity essential assembly complex. mutations no effect RO1 levels only very slight localization pattern The role RO11 movement N. hyphae remains unknown.
