Inhibition of α7‐containing nicotinic ACh receptors by muscarinic M1 ACh receptors in rat hippocampal CA1 interneurones in slices

摘要: Cys-loop ligand-gated nicotinic ACh receptors (nAChRs) and G protein-coupled muscarinic (mAChRs) are expressed on rat hippocampal interneurones where they can regulate excitability, synaptic communication cognitive function. Even though both nAChRs mAChRs appear to co-localize the same interneurones, it is not clear whether there crosstalk between them. We utilized patch-clamp techniques investigate this issue in CA1 slices under conditions transmission was blocked. The α7 nAChR-mediated currents were activated by choline, when activation of receptor preceded M1 mAChR subtype, amplitude responses significantly reduced a rapidly reversible voltage-independent manner, without any change kinetics responses. This mAChR-mediated inhibition through PLC-, calcium- PKC-dependent signal transduction cascade. These data show that functionally co-localized individual these particular inhibits nAChR information will help understand how cholinergic systems might be regulating neuronal excitability hippocampus manner has relevance for plasticity cognition.
