Celastrol induces apoptosis of human osteosarcoma cells via the mitochondrial apoptotic pathway

摘要: Celastrol is an active compound extracted from the root bark of Triptergium wilfordii Hook F., also known as 'Thunder God Vine'. It a well-known Chinese medicinal herb that was found to inhibit tumor cell growth and promote apoptosis in several lines. However, research into its effects on osteosarcoma still extremely limited. The present study undertaken determine effect celastrol viability cells furthermore, illuminate molecular mechanism celastrol-induced apoptosis. 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) colorimetric assay used evaluate following treatment with celastrol. apoptotic rate evaluated by flow cytometry using Annexin V-PE/7-AAD staining assay. Fluorescence microscopy detect morphological changes human U-2OS expression Bcl-2 family proteins, caspase-3, caspase-8, caspase-9, cytochrome c PARP measured western blotting. We significantly inhibited dose-dependent manner, particularly cells. Furthermore, we observed upregulated pro-apoptotic proteins Bax altered ratio Bax/Bcl-2, triggered mitochondrial pathway, resulting caspase-3 -9 activation cleavage. To conclude, results indicate inhibits proliferation cancer inducing via mitochondrial-dependent pathway.
