摘要: Oxymatrine, one of the most active components ethanol extracts from Sophora flavescens, is known for its potent antitumor activity both in vitro and vivo. However, mechanism action mediating cell apoptosis remains elusive. In this study, we investigated proliferation inhibitory apoptotic activities oxymatrine against human osteosarcoma MG-63 cells. The compound was found to markedly dose-dependently inhibit determined by 5-bromo-2-deoxyuridine incorporation. Oxymatrine also induced a dose- time-dependent manner as showed annexin V-FITC/PI double staining TUNEL assay. Furthermore, disruption mitochondrial membrane potential an up-regulation cleaved caspases-3, and-9 downregulation Bax/Bcl-2 evidenced oxymatrine-treated These proteins have been play pivotal role regulation apoptosis. conclusion, these observations indicate effective agent osteosarcoma. Moreover, appears exert anti-tumor stimulating caspase-triggered signaling pathway.
