Catharanthus roseus Aqueous Extract is Cytotoxic to Jurkat Leukaemic T-cells but Induces the Proliferation of Normal Peripheral Blood Mononuclear Cells.

摘要: Research on natural products has been widely used as a strategy to discover new drugs with potential for applications in complementary medicines because they have fewer side effects than conventional drugs. The aim of the present study was evaluate vitro cytotoxic crude aqueous Catharanthus roseus extract Jurkat cells and normal peripheral blood mononuclear (PBMCs). standardised vinblastine by high performance liquid chromatography (HPLC) determine cytotoxicity MTS [3-(4,5-dimethylthiazol-2-yl)-5(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium] assay. DNA fragmentation assay employed if cell death due apoptosis. results showed that induced at 24, 48 72 hours post-treatment time- dose-dependent manner. However, treated produced higher half maximal inhibitory concentration (IC50) values 2.55 μg/ml 2.38 μg/ml, respectively. In contrast, PBMC proliferation, especially after 24 treatment 1000 μg/ml. This result indicates C. differential inhibiting proliferation line promoting growth PBMCs. These data suggest may be applicable modulating transformed immune leukaemia patients.
