摘要: Genetic diseases and knockout mice stress the importance of matrix metalloproteinases (MMPs) in skeletal turnover. Our study aims at clarifying which MMPs are expressed by osteoclasts. Previous analyses this basic question led to conflicting reports literature. In present study, we used a variety approaches: PCR, Northern blots, Slot situ hybridization, immunohistochemistry. We analyzed osteoclasts culture as well native bone different locations compared mouse rabbit Osteoclasts express MMP-9 -14 all conditions, although variable extent, they able synthesize MMP-3, -10, -12, least under some circumstances. The induction given MMP is influenced its environment (e.g., osteoclast vs. bone, various sites within same bone) depends on species rabbit). show high amounts MMP-2 -13 protein presumably made large extent other cells, thereby documenting how proteinases nonosteoclastic origin may contribute activities giving insight why resorptive activity purified appears insensitive inhibitors. shows that confusion about osteoclastic literature reflects remarkable ability adapt their environment, required structural or functional diversity tissue. observations provide information needed for understanding emerging role controlling cell signaling resorption.
