High-affinity binding with specific peptides endows EuW10 a good luminescence probe for HPV E6 detection

摘要: Human papillomavirus (HPV) E6, acting as a major oncoprotein of high-risk sub-type, is known an ideal biomarker with diagnostic potential. Due to the bottleneck lacking effective, commercially available mono-clonal antibody (mAbs) for HPV16 we report new luminescence protocol in vitro E6 detection present study. By selecting inorganic cluster, Na9[EuW10O36]·32H2O (EuW10) that contains nine negative charges fluorescence probe, concentration low 0.28 μM was detectable buffer solution. For that, small peptide rich basic residues from used construct platform high-affinity interaction assay protein EuW10. Additionally, binding mechanism between EuW10 and revealed detail by time-resolved spectrometry ITC, while processes were monitored TEM images. Therefore, study not only provides practical support application polyoxometalate (POM) clusters biological probes HPV detection, but also significant early diagnosis HPV-induced cancers.
