Interpretation of Incomplete Reactions in tRNA Aminoacylation

摘要: A study has been performed in order to try explain some anomalies which are observed aminoacylation reactions: the extent of often depends upon experimental conditions, such as enzyme concentration or ionic strength. We have demonstrated that these can be explained, case yeast tRNAValII—valyl-tRNA synthetase system, by existence two reactions occurring beside reaction: spontaneous deacylation valyl-tRNAVal and a valyl-tRNA synthetase-catalyzed Val. Consequently, any plateau merely reflects an equilibrium between reaction reactions. Sometimes kinetics do not present but maximum value followed decrease with time. This explained additional degradation ATP into AMP PP1 presence tRNAVal valine. leads accumulation favouring reverse aminoacylation. The kinetic constants determined. Knowing constants, it is possible calculate theoretical values good agreement ones. These interpretations probably extended all tRNA systems.
