摘要: A procedure for designing the operation parameter space twin-column multicolumn countercurrent solvent gradient purification (MCSGP) process of therapeutic proteins is derived. This based on equilibrium theory, which assumes instantaneous conditions. As MCSGP allows protein separation with a linear modifier gradient, all equations are derived in terms covered distance as function concentration ion-exchange chromatography. All constraints, need to be fulfilled order obtain stable maximum yield and purity, described different parameters. For parameters within where constraints fulfilled, predicted. Additionally, boundary this region, optimal point buffer consumption productivity can found. Besides, presented design help analyze impact diff...
