摘要: An understanding of developmental processes requires knowledge transcriptional and epigenetic landscapes at the level tissues ultimately individual cells. However, obtaining tissue- or cell-type-specific expression chromatin profiles for animals has been challenging. Here we describe a method purifying nuclei from specific cell types animal models that allows simultaneous determination both profiles. The is based on in vivo biotin-labeling nuclear envelope subsequent affinity purification nuclei. We use to isolate muscle adult Caenorhabditis elegans mesoderm Drosophila melanogaster embryos. As case study, determined nucleosome occupancy affinity-purified C. muscle. identified hundreds genes are specifically expressed found these depleted nucleosomes promoters gene bodies relative other tissues.
