Kindling-induced changes in plasticity of the rat amygdala and hippocampus.

摘要: Temporal lobe epilepsy (TLE) is often accompanied by interictal behavioral abnormalities, such as fear and memory impairment. To identify possible underlying substrates, we analyzed long-term synaptic plasticity in two relevant brain regions, the lateral amygdala (LA) CA1 region of hippocampus, kindling model epilepsy. Wistar rats were kindled through daily administration brief electrical stimulations to left basolateral nucleus amygdala. Field potential recordings performed slices obtained from 48 h after last induced seizure, sham-implanted nonimplanted controls. Kindling resulted a significant impairment potentiation (LTP) both LA CA1, magnitude which was dependent on number prior stage V seizures. Saturation CA1-LTP, assessed repeated spaced delivery high-frequency stimulation, occurred at lower levels compared animals, consistent with hypothesis reduced capacity further strengthening. Furthermore, theta pulse stimulation elicited depression controls, whereas same protocol caused LTP rats. In conclusion, differentially affects magnitude, saturation, polarity LA, respectively, most likely indicating an activity-dependent mechanism context metaplasticity.
