摘要: The idea of “receptor mosaics” is that proteins may form complex and dynamic networks with respect to time composition. These have the potential markedly expand diversity specificity G protein-coupled receptors (GPCR) signaling, particularly in neural cells, where a few key been implicated many neurological psychiatric disorders, including addiction. Metabotropic glutamate type 5 (mGlu5) can complexes other GPCRs, adenosine A2A dopamine D2 receptors. mGlu5-containing reported striatum, brain region critical for mediating rewarding incentive motivational properties drugs abuse. and/or downstream interactions between divergent play roles addiction–relevant behaviors. Interactions mGlu5 GPCRs regulate conditioned effects as well drug-seeking influence striatal function, GABAergic output striatopallidal neurons glutamatergic input from corticostriatal afferents. Given their discrete localization, mGlu5-[non-mGlu5] receptor minimize off-target thus provide novel avenue drug discovery. therapeutic targeting receptor–receptor functional mosaics tissue specific or temporal manner (for example, sub-population “pathological state”) might reduce detrimental side otherwise impair vital functions.
