Quantitative evaluation of DNA hypermethylation in malignant and benign breast tissue and fluids

摘要: The assessment of DNA had demonstrated altered methylation in malignant compared to benign breast tissue. purpose our study was (i) confirm the predictive ability tissue, and (ii) use genes found be cancer tissue evaluate diagnostic potential hypermethylation nipple aspirate fluid (NAF) mammary ductoscopic (MD) samples. Quantitative specific (qMS)-PCR conducted on three specimen sets: 44 (CA) 34 normal (NL) specimens, 18 matched CA, adjacent (ANL) NAF 119 MD specimens. Training validation sets were analyzed determine optimal group for analysis. cytologic review also performed. Methylation CCND-2, p16, RAR-β RASSF-1a significantly more prevalent tumor than Receiver operating characteristic curve analysis an area under 0.96. For ANL four identified contained increased CA vs. tissue; samples higher frequency specimens from breasts with p16 RASSF-1a. In summary, i) routine quantitative is possible, ii) hypermethylated are samples, may useful assist early detection.
