CCL17/TARC and CCR4 expression in Merkel cell carcinoma.

摘要: Merkel cell carcinoma (MCC) is a rare, highly aggressive neuroendocrine skin cancer. In more than 80% of the cases, polyomavirus (MCPyV) causal factor. The oncogenic potential MCPyV mediated through its viral oncoproteins, large T antigen (LT) and small t (sT). To investigate role cytokines in MCC, PCR array analysis for genes encoding inflammatory receptors was performed on MCPyV-negative MCPyV-positive MCC lines, respectively. We detected an increased expression CCL17/TARC MKL2 line compared to MCC13 line. Transfection studies cells with LT plasmid, luciferase reporter plasmid containing promoter, exhibited stimulated promoter activity. Interestingly, ectopic upregulated early late activities cells. Furthermore, recombinant activated both mitogen-activated protein kinase NF-κB pathways. Finally, immunohistochemical staining human tissues showed strong receptor CCR4 LT-positive -negative MCC. Taken together, may be target therapy providing patients better overall survival outcome.
