摘要: A novel bioluminescence assay has been developed for measuring pyruvate within sections of snap-frozen tissue in a quantitative manner as well with spatial resolution on microscopical level. The was verified via HPLC and two independent photometric tests. makes it possible to determine concentrations cryosections the range 0–5.0 μmol/g (dry weight). Based analysis samples given concentrations, exhibits recovery deviation ≤15%. minimal detectable amount 0.02 pmol based 20 μm thick section an area 1 cm2. Combination already established imaging techniques ATP, glucose, lactate allows comprehensive characterization metabolic profile individual tumors. As redox state cancer cells can be critical efficiency irradiation number chemotherapeutics, are known have radical scavenger functions, we hypothesize that may used pretherapeutic lactate-to-pyruvate ratio which predict radiosensitivity malignancies.
