摘要: Mycoplasma hyorhinis has been shown to induce the secretion of tumor necrosis factor alpha (TNF-alpha) from monocytes. To identify molecules responsible for this activity, we separated sonicated M. lysate material by centrifugation at 100,000 x g into soluble (S) and particulate (P) fractions. The fractions were assayed TNF-alpha-inducing activity L929 bioassay. Both able TNF-alpha in roughly equal amounts. optimum dose both was 1 micrograms/ml. Proteinase K treatment either fraction eliminated suggesting that a protein component is involved induction. Phase partitioning Triton X-114 aqueous (A) detergent (D) phases showed composed 80% aqueous-phase proteins, while > 75% detergent-phase proteins. All four (SA, SD, PA, PD) release. Treatment with NaIO4 remove carbohydrate reduced inducing SA phase 80%, whereas other unaffected treatment.
