摘要: We show here that purified lipoarabinomannan (LAM) from Mycobacterium tuberculosis can cause the release of tumour necrosis factor (TNF) in vitro human blood monocytes and activated mouse peritoneal macrophages, production TNF vivo mice pretreated with Propionibacterium acnes, a potency comparable to lipopolysaccharide (LPS) Gram negative bacteria. Like LPS, LAM binds polymyxin B. confirmed its activity was distinct any contaminating LPS associated antigenic by affinity chromatography, using monoclonal antibody specific for LAM. Treatment dilute alkali greatly diminished TNF-inducing activity, suggesting omicron-acyl groups may be involved. When fractionated electrophoresis on SDS-Page blotted nitrocellulose, most capacity coincided bulk LAM, as estimated molecular weight activity. This modification Western blotting technique generally useful study macrophage-triggering molecules.
