摘要: A new method (enzyme-ligand immunoassay, ELIA) is described for the estimation of estrogen (ER) and progesterone (PR) receptors in microsamples human breast cancer tissue. The technique, based on nonisotopic measurement receptor-bound estradiol progesterone, involves three steps: (a) simultaneous saturation active with their respective authentic ligands, (b) heat treatment cytosol to release steroids from cognate before or after absorption dextran-coated charcoal, (c) both present by a modified competitive-inhibition enzyme immunoassay. useful range was 10-4000 pmol/L ER 6.5-1000 PR. correlation coefficient (r) between one-point Scatchard plot analysis 0.95 0.99 Comparison ELIA expected values radioligand binding assay (RLBA) results EORTC samples gave r = 0.88 PR, respectively. Further comparison RLBA commercial blind testing tissue 70 patients, good agreement 0.95-0.97 concordance 92.9-94.4% (cutoff, 15 pmol/g protein) against other two methods. were more disperse all methods PR estimation, correlating perhaps better immunoassay (r 0.90) at 89.4% (same cutoff value).
