摘要: Ca(2+)-induced exocytosis in chromaffin cells now seems to consist of at least two distinct steps:MgATP-dependent Ca(2+)-dependent priming the secretory apparatus, and MgATP-independent step that triggers (Bittner Holz, 1992). Recently we found a specific inhibitor myosin light chain kinase (MLCK), wortmannin, inhibits catecholamine release from digitonin-permeabilized cells, suggesting an implication MLCK mechanisms (Imaizumi et al., 1992b). To elucidate further mechanism exocytosis, studied effects wortmannin peptide (SM-1) corresponding pseudosubstrate domain on MgATP-dependent cells. permeabilized presence MgATP was inhibited by both SM-1 wortmannin. Inhibitory effect rate induced 10 microM Ca2+ much prominent later phase (1-10 min), although initial also decreased. strongly ATP-dependent without affecting ATP-independent all.
