The role of surface charge density in cationic liposome-promoted dendritic cell maturation and vaccine-induced immune responses

摘要: Cationic liposomes have emerged as a novel adjuvant and antigen delivery system to enhance vaccine efficacy. However, the role of surface charge density in cationic liposome-regulated immune responses has not yet been elucidated. In present study, we prepared series DOTAP/DOPC with different densities by incorporating varying amounts DOPC (a neutral lipid) into DOTAP lipid). The results showed that relied on density, might occur through ROS signaling. relatively high such 5:0 4:1 liposomes, potently enhanced dendritic cell maturation, generaion, uptake, well production OVA-specific IgG2a IFN-γ. contrast, low-charge 1:4 liposome, failed promote even at concentrations, confirming immunoregulatory effect is mostly attributable their density. Moreover, liposome suppressed anti-OVA antibody vivo. Overall, maintaining an appropriate crucial for optimizing enhancing efficacy liposome-based vaccines.
