Up-regulation of expression of selected genes in human bone cells with specific capacitively coupled electric fields

摘要: The objective of the described experiments was to determine electrical parameters that lead optimal expression a number bone-related genes in cultured human bone cells exposed capacitively coupled electric field. Human calvarial osteoblasts were grown modified plastic Cooper dishes which could be various fields. duration stimulation and response field, amplitude, frequency duty cycle all determined for each analyzed. Results indicated field 60 kHz, 20 mV/cm, 50% 2 h per day significantly up-regulated mRNA transforming growth factor (TGF)-β family (bone morphogenetic proteins (BMP)-2 -4, TGF-β1, - β2 -β3) as well fibroblast (FGF)-2, osteocalcin (BGP) alkaline phosphatase (ALP). Protein levels BMP-2 TGF-β1 also elevated. clinical relevance these findings context noninvasive treatment modality delayed union nonunion fracture healing is discussed. © 2014 Orthopaedic Research Society.
