摘要: Methylenetetrahydrofolate reductase (MTHFR) is an important enzyme involved in folate metabolism, which essential for DNA synthesis and methylation. Genetic variations the MTHFR gene seem to contribute a decreased activity of MTHFR, ultimately confer increased susceptibility cancer. As most extensively studied polymorphism, C677T polymorphism was shown cancer but results were inconsistent. The authors performed meta-analysis including 134 studies (46,207 cases 69,160 controls) address issue. Odds ratios (ORs) with corresponding 95 % confidence intervals (CIs) used assess association. Overall, significant elevated risk associated T-allele versus C-allele comparison (OR = 1.06, CI 1.02–1.11, P heterogeneity < 0.001), homozygote model (OR = 1.08, 1.01–1.17, < 0.001) dominant (OR = 1.05, 1.00–1.10, < 0.001). In stratified analyses, significantly risks indicated among Asians all genetic models except heterozygote model. Further analysis revealed that esophageal stomach This supports association between cancer, especially Asians. Additionally, more high-quality covariates responsible heterogeneity should be controlled obtain conclusive response about function
