Genetic alterations in cervical carcinomas: frequent low-level amplifications of oncogenes are associated with human papillomavirus infection.

摘要: The development of cervical carcinoma is closely associated with HPV infection. However, other genetic alterations also play an important role. In this study, we analyzed copy number several oncogene loci in a panel 84 tumors. Sixty-five (77%) tumors were DNA-positive, and most infected type 16 or 18 both. oncogenes studied include PIK3CA at 3q26.3, TERT 5p15.33, C-MYC 8q24, CCND1 11q13.3, ERBB2 17q21.2 locus region 20q13.2. Amplification 1 more genes was detected 55 (65%) cases using interphase FISH. amplified 43% tumors, followed by (33%), 20q13.2 (30%), (29%), (25%) (12%). Most showed low-level amplification 3–7 copies these genes, complex changes involving 3 occur frequently advanced stages. Increased protein expression c-erbB2 c-myc observed the corresponding gene amplification. Oncogene found often HPV-infected cases, particularly for TERT. These findings indicate that HPV-associated carcinomas bear frequent which may have critical biologic impact on progression uterine cervix. © 2002 Wiley-Liss, Inc.
