Protective effect of polyphenols in an inflammatory process associated with experimental pulmonary fibrosis in mice.

摘要: Polyphenols have been described to a wide range of biological activities, and many reports, published during recent years, highlighted the beneficial effects phenolic compounds, illustrating their promising role as therapeutic tools in several acute chronic disorders. The purpose study was evaluate, an already-assessed model lung injury caused by bleomycin (BLM) administration, resveratrol quercetin, well explore potential properties mango leaf extract, rich mangiferin, grape dihydroquercetin (DHQ), on same model. Mice were subjected intra-tracheal administration BLM, polyphenols administered oral route immediately after BLM instillation daily for 7 d. Treatment with resveratrol, quercetin DHQ inhibited oedema formation body weight loss, ameliorated polymorphonuclear infiltration into tissue reduced number inflammatory cells bronchoalveolar lavage fluid. Moreover, suppressed inducible nitric oxide synthase expression, prevented oxidative nitroxidative injury, shown nitrotyrosine poly (ADP-ribose) polymerase levels. degree apoptosis, evaluated Bid Bcl-2 balance, also polyphenol treatment. Finally, these natural products down-regulated cyclo-oxygenase-2, extracellular signal-regulated kinase phosphorylated expression NF-κBp65 translocation. Our findings confirmed anti-inflammatory BLM-induced damage, highlight, first time, protective exogenous mangiferin experimental pulmonary fibrosis.
