摘要: Several species of Aspergillus section Nigri, including potential mycotoxin producers, are common residents grape vineyards, but the relative population size individual throughout growing season is difficult to determine using traditional isolation and identification methods. Using a quantitative droplet digital PCR (ddPCR) method in combination with dilution plating, total Nigri populations proportions A. niger, welwitschiae, carbonarius, tubingensis were measured from vineyard samples without need for identifying fungal isolates. Grapes sampled two raisin vineyards (vineyards A B) at berry set, veraison, harvest, stages consecutive years. Plate counts showed that present on fruit increased set became larger component recovered both Results ddPCR analysis proportion carbonarius among four assayed later (harvest raisin) comparison earlier (berry veraison). Total plate not significantly different between either year. However, generally higher harvest than B. This coincided incidence levels ochratoxin B fruit. work demonstrates this useful tool culture-independent monitoring mycotoxigenic during production.
