β-Catenin mutations in liver tumors induced by 2-amino-3,4-dimethylimidazo[4,5-f]quinoline in CDF1 mice

摘要: Abstract Heterocyclic amines are potent mutagens and carcinogens formed in cooked protein rich foods. In this study, we screened liver tumors induced by 2-amino-3,4-dimethylimidazo[4,5-f]quinoline (MeIQ) CDF1 mice for β-catenin APC mutations other genetic alterations shown to occur human hepatocellular carcinomas (HCC), including the p53 H-ras genes, c-myc amplification E-cadherin promoter methylation. SSCP followed direct DNA sequencing revealed exon 2 of gene 16 (12.5%). Promoter methylation was detected one tumor MeIQ. There were no mutation cluster region gene, exons 5–8 or codons 12, 13 61 nor any These data indicate that except occasional disruption Wnt pathway through mutations, pathways involved development HCC differ significantly between cancer MeIQ, but do not rule out possibility heterocyclic constitute a carcinogenic risk factor humans.
