摘要: Purpose: Caffeic acid phenethyl ester (CAPE) is the main polyphenol extracted from honeybee propolis, which inhibits growth of several kinds tumor. This study aimed to assess inhibitory effect CAPE in nasopharyngeal carcinoma (NPC), evaluate synergistic action radiotherapy sensitivity NPC cell lines and further elucidate possible molecular mechanism involved. Materials methods: CCK-8 assay was used analyze proliferation ability. Colony formation clonogenic ability radio-sensitiveness cells by treatment. Wound-healing transwell were motility cells. The expression key molecules epithelial-mesenchymal transition (EMT) determined western blot analysis changes radiation measured colony-formation assay. cDNA microarray determine differentially expressed genes with without treatment, Gene Ontology enrichment gene function KEGG pathways determined. Cell cycle apoptosis detected flow cytometry analysis. Results: suppressed viability time- dose-dependently. It induced along decreased Bcl-XL increased cleavage PARP Bax. G1 phase arrest ower CDK4, CDK6, Rb p-Rb. migratory invasive EMT pathway. irradiation enhanced specifically inhibited nuclear factor κB (NF-κB) signaling pathway suppressing p65 subunit translocation cytoplasm nucleus. treatment chemotherapy radiotherapy. Conclusion: may inhibit metastasis but enhance radiosensitivity therapy inhibiting NF-κB could be a potential therapeutic compound for therapy.
