A Quantitative Reverse Transcriptase-Polymerase Chain Reaction Assay to Identify Metastatic Carcinoma Tissue of Origin

摘要: Identifying the primary site in patients with metastatic carcinoma of unknown origin can enable more specific therapeutic regimens and may prolong survival. Twenty-three putative tissue-specific markers for lung, colon, pancreatic, breast, prostate, ovarian carcinomas were nominated by querying a gene expression profile database performing literature search. Ten these marker candidates then selected based on validation reverse transcriptase-polymerase chain reaction (RT-PCR) 205 formalin-fixed, paraffin-embedded specimens originating from six other cancer types. Next, we optimized RNA isolation quantitative RT-PCR methods 10 applied assay to set 260 tumors. We built gene-based algorithm that predicted tissue an overall leave-one-out cross-validation accuracy 78%. Lastly, our demonstrated 76% when tested independent 48 samples, 37 which either known or initially presented as but subsequently resolved.
