摘要: In Colletotrichum lagenarium, which is the causal agent of cucumber anthracnose, PEX6 required for peroxisome biogenesis and appressorium-mediated infection. To verify roles peroxisome-associated metabolism in fungal pathogenicity, we isolated functionally characterized ICL1 C. encodes isocitrate lyase involved glyoxylate cycle peroxisomes. The icl1 mutants failed to utilize fatty acids acetate growth. Although Icl1 has no typical peroxisomal targeting signals, expression analysis GFP-Icl1 fusion protein indicated that localizes These results indicate occurs inside acid Importantly, contrast with pex6 form nonmelanized appressoria, formed appressoria were highly pigmented melanin, suggesting not essential melanin biosynthesis appressoria. However, exhibited a severe reduction virulence. Appressoria develop penetration hyphae host plant, invasion. addition glucose partially restored virulence mutant. Heat shock treatment plant also enabled lesions, implying infection defect mutant associated defense. Together requirement appressorial melanization, our findings suggest metabolic pathways play functional melanization subsequent invasion steps, latter step requires cycle.
