Detection of DNA hybridization by field-effect DNA-based biosensors: mechanisms of signal generation and open questions

摘要: Abstract We model theoretically the electrostatic effects taking place upon DNA hybridization in dense arrays immobilized on a layer of Au nano-particles deposited surface field-effect-based capacitive biosensor. consider influence separation charged analyte from sensor and salinity electrolyte solution, framework both linear nonlinear Poisson–Boltzmann theories. The latter predicts substantially weaker signals due to saturation that is main conclusion this paper. analyze how different physical parameters brushes affect magnitude signals. list includes fraction charge neutralization, length spatial conformations adsorbed molecules, as well discreteness charges. also examine effect Donnan ionic equilibrium lattices response. validity theoretical models contrasted against recent experimental observations detection via its intrinsic electric charge. sensitivity such biochemical sensing devices, their limit, efficiency are briefly discussed end.
