摘要: Azo dyes are widely used in the food, pharmaceutical, cosmetic, textile and leather industries. They can be reduced by azoreductases intestinal bacteria, liver cells skin surface microflora so that aromatic amines released. In this study an analytical system for determination of carcinogenic at picogram to femtogram level a cell culture assay evaluate toxicological effects azo is presented. For assays, commercial dye Resacor Blue 2F (Color Index: Direct 15), 3,3′-dimethoxybenzidine-based dye, was used. The released amine, 3,3′-dimethoxybenzidine, extracted with diethylether derivatized pentafluoropropionic anhydride analyzed capillary gas chromatography/mass spectrometry. addition, cytotoxicity on cultured kidney epithelial two hepatocyte lines (Hep G2 Chang liver) evaluated. purpose, were exposed 72 h varying concentrations 2F. results show inhibits proliferation much more than lines. As calculated from dose-response curves, EC50 (effective concentration reducing 50%) 40 μg/ml, whereas both 250 μg/ml. Higher 3,3′-dimethoxybenzidine found Hep supernatants; only small amounts supernatant. summary, it demonstrated supernatants, but did not accumulate within cells. interpretation results, cell-specific transport systems osmotic which contains several inorganic organic additives has considered.
