Genotype and Parental Combination Influence Efficiency of Cybrid Induction in Citrus by Electrofusion

摘要: Interspecific hybridizations by electrofusion of embryogenic callus cells from 'Seminole' tangelo ( Citrus reticulata Blanco x C. paradisi Macf.), 'Hazzara (Abohar)', or 'Ohta' ponkan Blanco) and leaf 'Lisbon' lemon limon (L.) Burm. f.) rough (C. jambhili Lush.), respectively, were performed. Electrofusion lemon, (Abohar)' resulted in 33, 43, 36 plants, respectively. Seven to 10 plants each combination selected randomly used investigate nuclear cytoplasmic genomes. Regenerated derived tan- gelo possessed the same restriction fragment pattern for rDNA as that mesophyll parents mitochondrial DNA (mtDNA) respective suspension parents, indicating they cybrids. In contrast, all resulting a between confirmed mtDNA analysis be somatic hybrids. The chromosome number sup- ported results Southern blot hybridization. suggest specific cell lines, parental combinations, both can increase efficiency inducing cybrids electrofusion.
