摘要: Cytoplasmic male sterility (CMS) is known to be controlled by mitochondrial genome in higher plants including Satsuma mandarin (Citrus unshiu Marc.). Citrus symmetric fusion experiments often produce diploid cybrids possessing nuclear DNA from the mesophyll parent and (mtDNA) embryogenic callus parent. Therefore, it possible transfer CMS as seedy citrus cultivars leaf one somatic cybridization. Herein, technique was adopted create for potential seedlessness transferring Marc. cv. Guoqing No. 1 (G1) two traditional Chinese cultivars, ‘Shatian’ pummelo (C. grandis (L) Osbeck) ‘Bingtang’ orange sinensis Osbeck). Flow cytometry analysis showed that 19 recovered G1 + ‘Bingtang’ 17 of 35 regenerated G1 + ‘Shatian’ were diploid. The remaining tetraploid. combinations confirmed true simple sequence repeat (SSR) cleaved amplified polymorphic (CAPS) analysis, with their corresponding mtDNA common suspension parent, G1 mandarin. identified hybrids G1. chloroplast (cp-SSR) revealed hybrid/cybrid most cases possessed either parental type, G1 +‘Shatian’ all embryoids analyzed (cpDNA) both parents. These results demonstrated we succeeded introducing into target through fusion.
