Cytoplasmic Expression of Pontin in Renal Cell Carcinoma Correlates with Tumor Invasion, Metastasis and Patients’ Survival

摘要: Renal cell carcinoma (RCC) is the most lethal of all genitourinary malignancies. Distant metastasis represents major cause death in patients with RCC. Recent studies have implicated AAA+ ATPase pontin many cellular activities that are highly relevant to carcinogenesis. In this study, we demonstrate for first time was up-regulated RCC, and plays a previously unknown pro-invasive role metastatic progression RCC through epithelial-to-mesenchymal transition (EMT) pathway. 28 pairs freshly frozen clear samples matched normal renal tissues analyzed by quantitative RT-PCR western blotting demonstrated than tissues. addition, immunohistochemistry used evaluate subcellular expression 95 patients, found overexpression cytoplasm positively correlated features, predicting unfavorable outcomes patients. Furthermore, vitro experiments show predominantly expressed lines, significant suppression migration invasion siRNA treated lines observed. Mechanistic depletion E-cadherin protein down-regulated vimentin protein, decreased nuclear β-catenin expression, suggesting involvement EMT induced progression. Together, our data suggest as potential prognostic biomarker provide new promising therapeutic targets clinical intervention kidney cancers.
