摘要: Two components of a receptor complex for IL-13, the IL-4R and low affinity IL-13-binding chain, IL-13Rα1, have been cloned in mice humans. An additional high binding chain IL-13Rα2, has described We isolated cDNA from thymus that encodes murine orthologue human IL-13Rα2. The predicted protein sequence IL-13Rα2 (mIL-13Rα2) 59% overall identity to is closely related subunit, IL-13Rα1. genes both mIL-13-binding chains map X chromosome. A specific interaction between mIL-13Rα2.Fc IL-13 was demonstrated by surface plasmon resonance using BIACORE instrument. Ba/F3 cells were transfected with mIL-13Rα2 expressed 5000 molecules per cell bound single K d 0.5 1.2 nM. However, these did not proliferate response IL-4 dose unaffected concentrations IL-13. In contrast, expression mIL-13Rα1 resulted sensitive proliferative Consistent its lower IL-13Rα1.Fc 100-fold less effective than IL-13Rα2.Fc neutralizing vitro. These results show are functionally equivalent predict distinct roles each polypeptide IL-13R formation modulation signal transduction.
