MicroRNA-21 promotes oral cancer invasion via the Wnt/β-catenin pathway by targeting DKK2.

摘要: MicroRNA-21 (miR-21) is overexpressed in a wide variety of cancers and has been related to cellular proliferation, apoptosis, invasion; however, the function miR-21 unknown oral tongue squamous cell carcinoma (OTSCC). The purpose this study was examine expression OTSCC, correlate it with clinicopathological factors, investigate its contribution OTSCC invasion. MiR-21 79 primary OTSCCs evaluated using locked nucleic acid situ hybridization, correlation examined factors. To determine target, we searched for molecular genes involved tumor invasion commonly cited prediction program miRanda. In an line, SCC25 cells, further whether contributes invasiveness by blocking specific knockdown LNA probe confirmed direct target Matrigel assay Western blotting. overexpression detected 60 cases (75.9 %) correlated pattern (P = 0.016). We selected DKK2 as Wnt/antagonist significantly DKK2-/β-catenin- immunohistochemical phenotype. Knockdown decreased potential cells up-regulated DKK2. It found that associated promotes via Wnt/β-catenin pathway targeting vitro. These results suggest may be therapeutic treatment.
