摘要: Abstract Neurotrophins have been demonstrated to play important roles in the development and functioning of nervous system. This family proteins consists four homologous members mammals: NGF, BDNF, NT-3, NT-4/5. A new member, called NT-6, was recently cloned from platyfishXiphophorus maculatus.This protein shares closer structural relationship NGF than other neurotrophins, but contains a characteristic insertion 22 amino acids that constituted heparin-binding domain. Here we report cloning novel neurotrophin fishCyprinus carpio(carp), which shared about 66% acid identity toXiphophorusNGF NT-6. The neurotrophin, designated NT-7, possesses characteristics common all known such as presence six conserved cysteine residues flanking sequences. In addition, there is an 15 at position corresponding observed for neurotrophic activity NT-7 by its ability promote neurite outgrowth neuronal survival chick dorsal root ganglia. Phosphorylation assay various Trk receptors overexpressed fibroblasts suggested could activate TrkA not TrkB or TrkC. Northern blot analysis revealed predominantly expressed peripheral tissues, though weak expression also detected brain. Like this might represent yet another NGF-like lower vertebrates.
