Synthesis of prostaglandins by pig blastocysts cultured in medium containing estradiol or catechol estrogen.

摘要: Two experiments were conducted to determine the effects of 2-hydroxy-estradiol-17 beta (2-OH-E2; 0, 50 and 100 microM) estradiol-17 (E2; 25 on prostaglandin (PG) E PGF2 alpha synthesis by day-10 pig blastocysts (day 0 is first day estrus). Blastocysts incubated in a modified Krebs-Ringer bicarbonate medium, supplemented with bovine serum albumin (4 mg/ml) vitamins amino acids (essential nonessential) Minimum Essential Medium (without phenol red or antibiotics). The incubations at 39 degrees C for three 2-h periods; second third periods included an E2 catechol estrogen treatment. Release into culture medium decreased (p less than 0.001) linearly increasing concentrations 2-OH-E2 both periods. PGE was not affected 2-OH-E2, therefore increased 0.06) PGE:PGF2 alpha. When added release 0.01) during also 0.05) period 2, but did alter Content PGs recovery 10% released vitro. Therefore, these studies demonstrate primary forms Catechol estrogens may inhibit PG modify establishment pregnancy pigs.
