Computerized quantitative analysis of coomassie-blue-stained serum proteins separated by two-dimensional electrophoresis.

摘要: Two-dimensional electrophoresis in combination with Coomassie Blue staining was refined for use as a quantitative method. Microcomputer software developed the IBM AT and compatible computers analyzing gels. To test method to determine its usefulness simultaneous measurements of 28 human serum proteins, we measured each protein relative single standard (bovine albumin) polymerized at different concentrations calibration scale, rather than using individual standards. All samples were analyzed triplicate. We evaluated calibration, linearity response, recoveries, units, within-run CV, between-run CV. The five isoforms apolipoprotein A-I from 16 healthy donors isoform ratios determined. presented here should prove useful diagnosis non-urgent disease states analysis relation disease; it also be applicable assays proteins other fluids tissues.
