摘要: The anti-tumor mechanisms of local LAK cell therapy are difficult to study in vivo. We describe a method vitro the action cells against three-dimensional tumor tissue. Spherical aggregates (spheroids) grown from human glioma lines H-2 and U-251 were labeled with 51Cr then incubated for up 24 h cells. After incubation, most spheroids still macroscopically identifiable, measured reduction volume did not correlate extent damage. infiltrated into spheroid tissue slowly as frontier which explains why specific release was clearly slower than corresponding single suspensions. area at 1 2 very thin but by 8 12 consisted already several layers. Most became totally 16 whereas infiltration usually remained peripheral. In accordance different infiltration, more sensitive spheroids: E/T ratio 10:1 mean 63 36%, respectively. A exposure released approximately over spheroids. model can be used infiltrative capacity cytotoxicity tissue, may help find an optimal mode therapy, i.e., proper combination lymphokines cells, timing their administration.
