A model of cell activation and desensitization by surface immunoglobin: The case of histamine release from human basophils

摘要: We present a model for the control of immunoglobulin E (IgE)-mediated histamine release from human basophils. suggest that there is calcium gating factor which interacts with crosslinked IgE to form short-lived open channel. After formation channel activated rapidly decays an inactive form. It loss active causes basophil desensitize nonspecifically. propose molecules are deactivated by mechanism, such as endocytosis or shedding, independent mechanism inactivates factor. This functional leads specific desensitization. The mathematical formulation explains relationship and nonspecific desensitization amount on surface; why two types antigen excess inhibition; between inhibition desensitization; why, fixed concentration, increasing concentration cell surface increases until optimal reached, then decreases release; predicts effects changing external will have dose response curve; cause curve undergo transition single maximum maxima.
