摘要: The i.v. inoculation of parental spleen cells into unirradiated adult F1 hybrid mice results in a graft-versus-host reaction (GVHR). In the strain combination B10D2 --> (B10.BRx B10.D2) F1, this is associated with thymic injury and transient but profound cellular immune deficiency. We further analysed status these 60 days after GVHR induction. Phenotypic studies showed that were repopulated lymphocytes resulting high degree chimerism (85%). At time, looked healthy recovered normal cytotoxic T cell response (CTL) against allogeneic cells. GVH chimeric splenocytes unresponsive mixed lymphocyte culture (MLC), exhibited anti-F1 CTL reactivity. also reactivity vivo. unable to induce injection new specifically rejected bone marrow (BM) lethal irradiation. Grafting neonatal thymus prevented rejection BM restored alloreactivity. It concluded state induced by split tolerance radiosensitive thymic-dependent mechanism involved maintaining self-tolerance mice.
