摘要: Abstract The fluorescein leakage test (FLT) provides information on the effects of xenobiotics impermeability (gate function) epithelial cell monolayers, and their recovery after exposure. aim this study was to assess validity in ocular safety assessment surfactant-based products with various irritant potencies. Madin-Darby canine kidney cells were grown confluency microporous membranes exposed for 15 min increasing concentrations substances. Damage evaluated by measuring amount Na-fluorescein that passed through monolayer 30 min, starting just Recovery assessed 4, 24, 48 72 hr later. For each sample time point, amounts substance produced 10% 20% (FL10 FL20) compared a cell-free control calculated. 43 samples, FL20 values ranged from 0.65 1000 mg/ml. These increased or decreased according substance. In particular, monolayers showed very different recoveries exposure anionic cationic substances similar initial values. vitro data correlated well historical Draize vivo (Spearman's ϱ > 0.90). FLT is therefore useful as complement other methods evaluation cosmetics.
