摘要: PURPOSE. The ocular toxicity of various compounds is typically determined by the Draize eye test, which has been criticized in past for its lack reproducibility and cruelty associated with harsh testing conditions animals. In this study, a liposome-based assay was developed estimating surfactants. METHODS. release calcein dye from liposomes induced interactions surfactants studied correlated to scores. First, liposome conducted at identical concentrations Next, mechanistic geometric considerations were used determine appropriate surfactant concentration that should be assay. RESULTS. Correlations between percentage released scores drastically improved after chosen based on CMC/200, where CMC critical micelle With choice concentration, excellent correlations obtained three separate sources two different loadings (Pearson = 0.99, 0.82, 0.78, 0.74; Spearman 0.94, 0.79, 0.85). Subsequently, toxicities six nonionic surfactants, Brij 700, -58, -56, -78, -97, -98 shown minimal proposed correlations.
