Effects of the combination of metyrapone and oxazepam on cocaine craving and cocaine taking: a double-blind, randomized, placebo-controlled pilot study.

摘要: Although cocaine dependence affects an estimated 1.6 million people in the USA, there are currently no medications approved for treatment of this disorder. Experiments performed animal models have demonstrated that inhibitors stress response effectively reduce intravenous self-administration. This exploratory, double-blind, placebo-controlled study was designed to assess safety and efficacy combinations cortisol synthesis inhibitor metyrapone, benzodiazepine oxazepam, 45 cocaine-dependent individuals. The subjects were randomized a total daily dose 500 mg metyrapone/20 oxazepam (low dose), 1500 (high or placebo 6 weeks treatment. outcome measures reduction craving associated use as determined by quantitative measurements metabolite benzoylecgonine (BE) urine at all visits. Of subjects, 49% completed study. combination metyrapone well tolerated tended use, with significant reductions several time points when controlling baseline scores. These data suggest further assessments ability support abstinence warranted.
