摘要: In metazoa, inductive cell-to-cell interactions play crucial roles, both in embryonic development and homeostasis. Many such are mediated by RTKs their soluble or surface-bound ligand growth factors. Type 1 RTKs, also called the ERBB HER family, comprises four proteins, which bind factors of EGF neuregulin families. The ERBB/HER proteins not only for determination several epithelial, glial, muscle, neuronal lineages, but they key players pathological processes, as malignant transformation. Multiple mechanisms underlay driver supportive functions tumors, include receptor overexpression, receptor’s point mutations, internal deletions, well autocrine loops, meaning that a cell initiates its own proliferation through secretion. Although all receptors share similar domain architecture, ERBB2/HER2 binds with no known ERBB3/HER3 harbors barely active kinase domain. Like other signal downstream ERK/MAPK PI3K/AKT pathways. However, characterized extensive receptor-receptor interactions, functioning preferred heterodimer partner. recent years, ability to redirect signals means network plasticity is emerging an important way developed tumors evade pharmacological interceptors aimed at specific nodes network.
