摘要: The liver cell adhesion molecule (L-CAM) and N-cadherin or adherens junction-specific CAM (A-CAM) are structurally related surface glycoproteins that mediate calcium-dependent in different tissues. We have isolated characterized a full-length cDNA clone for chicken used this to transfect S180 mouse sarcoma cells do not normally express N-cadherin. transfected (S180cadN cells) expressed on their surfaces resembled with L-CAM (S180L at confluence they formed an epithelioid sheet displayed large increase the number of gap junctions. In addition, S180cadN cells, like S180L accumulated cellular boundaries where it was colocalized cortical actin. were seen sites junctions but restricted these areas. Adhesion mediated by either inhibited treatment cytochalasin D disrupted actin network cells. Despite known structural similarities, there no evidence interaction between Doubly (S180L/cadN) also sheets. both areas contact presence antibodies CAMs required disrupt sheets Studies using divalent localize each perturb distributions indicated same interactions molecules. These data suggest Ca(++)-dependent likely play critical role maintenance epithelial structures support model segregation binding. They provide further so-called precedence hypothesis proposes expression homophilic binding necessary formation junctional epithelia.
