摘要: Abstract Aims The present study aimed to explore the preventive or therapeutic effect of peri-operative pregabalin treatment on memory deficits and related hippocampal inflammation following surgery in aged rats. Main methods Aged rats underwent abdominal sham surgery, were then divided into 2 groups, either early late treatment. Fourteen days after cognitive function was assessed using novel object recognition test, followed by measurement cytokines voltage-dependent calcium channel α2δ subunit (CACNA2D1). parabiotic experiments determined whether humoral neuronal pathway involved neuroinflammation development surgery. effects LPS-induced cytokine release from microglia also evaluated. Key findings Early treatment, which administered pre-operatively continued for 3 7 days prevented decreased pro-inflammatory levels. In contrast, no beneficial observed when post-operative period. levels CACNA2D1 did not change under any experimental condition. data cross-circulation (parabiosis) indicated that may induce via a neural transmission periphery brain. ex vivo further demonstrated had microglia. Significance Our highlight reveal during period can prevent It is likely this occurs through peripheral central neuro-immune interaction rather than direct anti-inflammatory effects.
