Liquid chromatography/mass spectrometry of conjugates and oxidative metabolites of indole‐3‐acetic acid

摘要: Indole-3-acetic acid and some of its hydroxylated derivatives amide ester-linked conjugates were analysed by high-performance liquid chromatography/mass spectrometry (HPLC/MS). The effluent from a capillary reversed-phase HPLC column was introduced into the ion source double-focusing mass spectrometer via frit interface, ions generated fast atom bombardment with 1% glycerol in mobile phase acting as matrix. Full-scan positive spectra obtained 1–2 ng non-polar 100 polar indoles. both indole-3-acetic contained dominant molecular ion, adducts an m/z 130 quinolinium ion. oxidized derivatives, such oxindole-3-acetic 5-hydroxy-indole-3-acetic acid, characterized presence strong response at 146, representing oxygenated absence 130. Further oxidation products 7-hydroxy-oxindole-3-acetic yielded containing intense 162 hydroxy-2-oxoquinolonium ion. The HPLC/MS system used to identify indole-3-acetylalanine purified extract elongating shoots Picea abies. structure elucidation based on spectrum, daughter accurate determination high-resolution spectrometry.
