Isolation of monoclonal antibodies that recognize the transforming proteins of avian sarcoma viruses.

摘要: Thirteen clones of hybrid cells which synthesize antibodies directed against the Rous sarcoma virus (RSV) transforming protein, pp60src, were isolated. Mouse myeloma fused with spleen from mice that had been immunized purified pp60src bacterial recombinants direct synthesis RSV src gene. The hybridomas survived selection medium screened by immunoprecipitation 32P-labeled lysates RSV-transformed cells. Monoclonal produced subclones derived 13 recognized encoded Schmidt-Ruppin and Prague strains cellular homolog pp60src. Antibody clone 261 a high affinity for viral yes gene product, 443 463 proteins viruses containing related genes fps ros. Several other low yes, fps, ros products could be detected in vitro phosphorylation after monoclonal antibody. All allowed casein an immune complex-bound reaction.
